If you using PHD2 ( which I use ) the answer is no
But if you rotate it more than 1 degree between sessions then you will have to re calibrate if you run auto calibration
I usually line up my USB socket on the camera to the top, in line with my guide scope adjust knob ( this orientation works best for my USB cable ) however I never take my camera out of the guide scope so it’s stays in the same orientation all the time )
I use a ZWO ASI 120MM- S and an Orion 50mm mini guide scope with helical focuser
