Calibration won't help if the readout noise is higher than the signal, which with really faint stuff could be the issue with shorter subs.

I think the time taken during the read out in proportion of the exposure time is very negligible in the end. It's not like you're going to miss that many photons
The SNR is the real issue for faint stuff.
