Cleaning sensor of DSLR

I have some annoying dust motes showing up in images with this camera. I know I can probably get by with flats but would like to remove the source of the problem anyway.
The Canon has a sensor clean routine but doesn't seem to do much. What about opening a Bulb exposure and having a few puff of air via a puffer bulb or even some canned air. Thoughts?
